Glycine for Sleep: The Evidence

An amino acid, a temperature mechanism and what the research shows.

Glycine is an amino acid that has been studied for its possible role in the body's natural wind-down at bedtime. Here is the mechanism and a measured look at the research.

What Is Glycine?

Glycine is the smallest amino acid and one of the building blocks the body uses to make protein. It is found naturally in protein-rich foods, particularly collagen-rich cuts of meat, bone broth and gelatine, and the body can also make it. Beyond its structural role, glycine acts as a signalling molecule in the central nervous system, where it works as an inhibitory neurotransmitter. That dual identity - a dietary amino acid that also influences nerve signalling - is why glycine has drawn interest in the context of relaxation and sleep. It is the same glycine that pairs with magnesium in magnesium glycinate, a form people often choose for evening use.

The Thermoregulation Mechanism

The most studied explanation for glycine and sleep involves body temperature. Falling asleep is associated with a natural drop in core body temperature, achieved partly by sending more blood flow to the hands and feet so heat is released. Research suggests glycine promotes this peripheral blood flow and supports the lowering of core temperature around bedtime, which aligns with the body's own sleep-onset signals. Glycine also acts on receptors in the brain, including in regions involved in regulating the sleep-wake cycle. These are mechanistic findings from the scientific literature that describe how the amino acid behaves, rather than a promise about any particular outcome.

What the Research Has Found

Several small studies have examined glycine taken before bed. Work by Bannai and colleagues (2012) explored glycine's effects on sleep and reported changes consistent with the thermoregulation mechanism, while earlier research by Inagawa and colleagues (2006) looked at subjective sleep quality in people with mild sleep complaints after an evening dose of around 3 g. These trials are small and exploratory, so they describe a promising direction in the literature rather than settled proof. Reported amounts in this research typically sit in the region of 3 g taken before bed. As always, responses differ between individuals, and study findings are not a guarantee of personal results.

Glycine Alongside Magnesium

Glycine appears most often in sleep products as part of magnesium glycinate, where it serves both as a carrier that aids absorption and as a compound with its own area of research interest. This is one reason glycinate is a popular form for an evening routine. It is worth being clear that the glycine content of a typical magnesium glycinate serving is modest and not the same as the higher amounts used in dedicated glycine sleep studies. Think of the two as complementary rather than interchangeable.

Getting Glycine From Food

If you would rather increase glycine through diet, the richest sources are collagen and gelatine - found in bone broth, slow-cooked cuts with connective tissue, and gelatine-based foods - along with protein-rich foods generally. A varied diet that includes these foods contributes glycine alongside other nutrients. For most people, food first is a sensible approach, with supplements considered only as a top-up within a balanced overall intake.

Using Glycine Sensibly

Glycine is generally well tolerated, but it is a supplement and not a medicine, and it is not a treatment for insomnia or any other condition. If you are considering it, treat it as one optional element within a wider routine that prioritises consistent timing, a wind-down period and limited evening light. People who are pregnant or breastfeeding, who take regular medication, or who have an ongoing health condition should check with a GP or pharmacist first. If sleep problems persist, seek proper assessment rather than relying on supplements alone.
